The relationship between histological grade, oestrogen receptor status, events and survival at 8 years in the NATO ('Nolvadex') trial

Abstract: Summary A pathological review was carried out on 600 patients with breast carcinoma entered into the 'Nolvadex'* Adjuvant Trial Organisation (NATO) study. The tumours were graded histologically and these results were compared with the oestrogen receptor (ER) status of the tumours, the numbers of recurrences and the length of survival of the patients. It was found that histological grading was predictive both in terms of events and survival, and correlates significantly with oestrogen receptor status; within hi… Show more

“…A significant proportion of all breast tumours (35-40%) are ER negative, and the incidence of ER-negative tumours increased 22-27% over the period [1974][1975][1976][1977][1978][1979][1980][1981][1982][1983][1984][1985] (Glass and Hoover, 1990). When compared with ER-positive tumours, ER-negative tumours are frequently more poorly differentiated (Singh et al, 1988), proliferate more rapidly (Meyers et al, 1977;Jonat and Maat, 1978) and the patients have a poorer prognosis (Shek and Godlophin, 1989). We now describe the isolation of the first ascites model of human breast cancer (MDA435/LCC6).…”

“…27 Low ER and PR content is associated with poor response to hormone therapy, decreased tumorfree intervals and survival, and increased tumor grade in humans. 2,17,38,39 In domestic cats, lower numbers of ER or PR have been reported in malignant mammary tumors than in benign tumors, 22,27,33 and in 1 study, PRs were present in a group of ER-negative feline mammary gland carcinomas. How MGA affects ER and PR in zoo felid mammary glands, whether steroid hormone receptors are expressed in mammary carcinomas from MGA-treated zoo felids, and whether receptor expression correlates with tumor behavior have not been investigated.…”

“…Mammary carcinomas in domestic cats have comparable aggressive features, high tumor grades (II or III), and evidence of metastasis at the time of diagnosis. 5,28 These features are associated with decreased survival in domestic cat 5,20,28 and human 2,4,38,39 mammary cancer patients and are likely also important prognostic indicators in zoo felids. In domestic cats, the advanced status of these cancers at the time of diagnosis may be due in part to delayed owner observation.…”

“…These studies included comparisons of meiphalan (Rubens et al, 1983), CMF (Richards et al, 1990) and tamoxifen (Singh et al, 1988) (Camplejohn et al, 1989;Camplejohn, 1992). Briefly, SOpm sections were dewaxed and taken to 50% alcohol overnight using a Histokinette tissue-processing machine.…”
