The relationship between reactive oxygen species, DNA fragmentation, and sperm parameters in human sperm using simplified sucrose vitrification with or without triple antioxidant supplementation

Abstract: Objective: This study examined whether the addition of triple antioxidants (3A)—10 µM acetyl-L-carnitine, 10 µM N-acetyl-L-cysteine, and 5 µM α-lipoic acid—in freezing-thawing medium during human sperm cryopreservation using the sucrose vitrification (SuV) and liquid nitrogen vapor (Vapor) techniques could improve post-thaw survival of spermatozoa. Methods: We analyzed 30 samples from healthy human sperm donors. Each sample was allocated into one of five groups: (1) fresh control, (2) SuV, (3) SuV+3A, (4) Vapo… Show more

“…Last, we utilized frozen sperm samples for DNA extraction. We acknowledge that frozen sperm may exhibit lower DNA integrity, viability, and motility values compared to fresh sperm [ 74 , 75 ]; however, it is challenging to preserve and manage fresh sperm over the long term; and there is limited information available regarding the effects of cryopreservation on epigenetic modulation in sperm, and existing studies present contradictory results [ 75 ]. In the future studies, we intend to investigate the impact of the cryopreservation process on sperm DNA methylation.…”

“…El botón espermático se resuspendió en 1 mL de PBS antes de cuantificar por citometría de flujo en un FACSCalibur (Becton Dickinson). Se utilizó una longitud de onda de excitación de 488 nm y longitud de onda de emisión de 520 nm; se contaron, al menos 10,000 eventos por muestra y se determinó la media de fluorescencia [90,91] utilizando el sistema de cómputo Flowing Software, versión 2.5.1.…”
