The role of electrophoresis in disease biomarker discovery

Abstract: There has been increased activity in the last few years in the search for disease markers using fractionation of complex biological fluids combined with MS. While electrophoretic and chromatographic separations have played a major role in this endeavor, this manuscript is limited to a review of electrophoretic methods that have been established for disease biomarker discovery. These methods include 2-DE, difference gel electrophoresis (DIGE), and CE. We define what constitutes a biomarker, identify the steps r… Show more

“…Depleted sera are then either separated by 2D gel electrophoresis (DIGE) that allows comparative analysis of three different samples in a single gel, or by tryptic digests of serum proteome fractions that have been labeled with so-called isobaric tags (iTRAQ technology), permitting parallel analysis of up to eight serum samples. [91][92][93] Identification of the in gel trypsin digested proteins (DIGE) or of the tryptic iTRAQ peptides is done by MS. The power of these technologies lies in a quantitative comparison of an identified peptide or protein in three (DIGE) or eight (iTRAQ) serum samples.…”

“…Throughout the last decade, CE has been established as a reliable tool in biomarker discovery [5] and anticancer research, especially in the analysis of antitumor drugs based on transition metal complexes [6,7]. It has been applied to investigate the stability of novel compounds but also the interactions of drugs with biomolecules, such as human serum proteins [8][9][10][11][12][13][14][15] and nucleotides [16][17][18][19][20][21], elucidating possible metabolic and reaction pathways of the drug candidates [6,7,16], notably always working under simulated physiological conditions with regard to pH, temperature, aqueous solutions, and/or buffer composition.…”
