The Role of Intrinsic Pathway in Apoptosis Activation and Progression in Peyronie’s Disease

Abstract: Peyronie's disease (PD) is characterized with formation of fibrous plaques which result in penile deformity, pain, and erectile dysfunction. The aim of this study was to investigate the activation of the intrinsic apoptotic pathway in plaques from PD patients. Tunica albuginea from either PD or control patients was assessed for the expression of bax, bcl-2 and caspases 9 and 3 using immunohistochemistry and by measurement of apoptotic cells using TUNEL assay. Bax overexpression was observed in metaplastic bone… Show more

“…On the other hand, in the case of extrinsic apoptosis, Caspase 8 is activated by signaling through death receptors and it can further activate Casp3, which is the common player in both apoptotic pathways. Moreover, Caspase 8 can cleave Bid and this event will further activate the mechanism of the intrinsic pathway [52,53].…”

“…BAX increases the permeability of the outer mitochondria membrane which leads to an e ux of cytochrome c. Cytochrome c then binds to Apaf-1 and initiates the initiator caspase 9 and this in turn initiates executioner caspases, such as caspases 3, 6, and 7. Caspase-3 cleaves PARP which leads to DNA fragmentation and then apoptosis [24]. To verify the speculation, we conducted qRT-PCR and evaluated its effect on apoptotic markers, caspase-3, and PARP.…”

“…The induction of apoptosis by the activation of caspase-9 involves intrinsic or mitochondrial pathway of apoptosis. While caspase-8 plays a major role in the extrinsic pathway of apoptosis (Loreto et al, 2014). Activation of caspase-9 and caspase-8 further activates effector cas-3, cas-6 or cas-7 (Shi, 2002;Yuan and Akey, 2013;Ouyang et al, 2012).…”
