The specificity and genetic background of the rye (Secale cereale L.) tissue culture response

Abstract: Rye is one of the most important crops in Eastern and Northern Europe. Despite the numerous beneficial features of rye, its annual production decreases successively which correlates with the lack of progress in its breeding compared with other cereals. Biotechnological methods could effectively improve the breeding of rye. However, their application is highly limited by the absence of an efficient procedure for plant regeneration in vitro, since rye is one of the most recalcitrant cereals with regard to the ti… Show more

“…For instance, maize contains 23 JAZ proteins, which is nearly twice as many as tobacco [157,158]. This suggests involvement in other hormone signaling pathways or abiotic stress tolerance [387]. Furthermore, the NAC transcription factor RIM1, a negative regulator of JA biosynthesis in rice, has not yet been identified in Arabidopsis and may thus be specific to monocots [387,388].…”

“…In Arabidopsis , specific HATs or HDACs are known to acetylate or deacetylate certain lysine residues (Figure 1) such as members from GNAT/MYST class - AtHAG1/AtGCN5 acetylates H3K9, H3K14, and H3K27, AtHAM1/2 acetylates H4K5, and AtHAG3 acetylates H3K14. On the other hand, HDACs include (1) HD2 (HISTONE DEACETYLASE 2), (2) SIR2 (SILENT INFORMATION REGULATOR PROTEIN 2), and (3) HDAs (HISTONE DEACETYLASE) class (Class I – III) [2224]. Amongst them, AtHDA6 and AtHDA19/HD1 from the HDAs class I are the best-studied HDACs which are shown to be involved in removal of a few of the acetylation marks on H3 and H4 tails [2527].…”

“…The biggest number of OTLs connected with AR/100A were localized in subgenome R and the positive effect originated from 'Modus' allele. It could be connected with general recalcitrance in regard to the in vitro culture response in rye (Ma et al 2003;Ma and Pulli 2004;Targońska et al 2013). Generally problems associated with rye anther and microspore culture are poor embryogenic callus induction, low green plant regeneration and high proportion of albinos (Ma and Pulli 2004).…”
