The structural modification of natural products for novel drug discovery

Abstract: Throughout history, natural products (NPs) have provided a rich source of compounds that have wide applications in the fields of medicine, health sciences, pharmacy and biology. Although naturally active substances are good lead compounds for the discovery of new drugs, most of them suffer from various deficiencies or shortcomings, such as complex structures, poor stability and solubility. Therefore, structural modification of NPs is needed to develop novel compounds with specific properties. Areas covered: Th… Show more

“…In Europe and the United States, 25% of prescriptions contain at least one extract from higher plants or compounds. In Germany, pure natural products are considered as drugs rather than food supplements and are covered by medical insurance (Yao et al, 2016a). In short, pure natural products in foreign countries have a good application base and a wide range of markets.…”

“…Traditional Chinese medicine is one of the promising strategies in treating AP due to its multi-targeting and lower side effects (Yao et al, 2016a). Despite massive investments in TCM research and development, there has been no significant increase in the number of new drugs approved or translated for clinical use.…”

“…It lies in structure-activity relationship (SAR)-oriented synthesis. 7,8 Formulation of SARs provides information of essential functional groups to guide practical structure optimization of parental compounds, 9 which further benets lowering side effects and improving efficacy. Therefore, the study of SARs of lead compounds is considerably vital in drug discovery.…”

“…Although naturally active substances are good material for the development of new drugs, most of them suffer from various deficiencies or shortcomings, such as complex structures, poor stability, or solubility. Chemical modification of plant compounds increases the activity or selectivity of antibiotics, improving their stability or physico-chemical properties [49,50].…”
