SummaryThe nucleic acid of infectious bronchitis virus (IBV), like that of other enveloped viruses, consists of discontinuous single stranded RNA. However, unlike many other viruses, there is extreme heterogeneity in the sizes of the I~NA fragments, as revealed by centrifugation in sucrose gradients or electrophoresis in polyacrylamide gels. Two principal classes of I~NA fragments are present: ~) A larger class comprising 74.9--85.4 per cent of total RNA and consisting of fragments having molecular weights ranging from 0.5 • 106 to considerably greater than 3.0 • 106 da]tons and, b) A smaller class comprising 9.1--19.7 per cent of total I~NA with the size approximately that of ribosomal r S I~NA.All IBV RNA's were fully susceptible to ribonuclease and had a buoyant density in caesium sulphate identical to that of tobacco mosaic virus RNA. :No difference in the R:NA profile for IBV was observed from the use of different methods of virus purification. The single-stranded R:NA's of poliovirus and tobacco mosaic virus remained undegraded after preparation in the presence of IBV.

“…The earlier of the two noted that the recognition of coronaviruses as a separate virus family occurred in the 1960s, in the wake of the discovery of several new human respiratory pathogens, certain of which, it was realized, appeared highly similar to the previously described avian infectious bronchitis virus (IBV) and mouse hepatitis virus (MHV) (Almeida and Tyrrell, 1967). These latter viruses had a characteristic morphology in negative-stained electron microscopy, marked by a "fringe" of surface structures described as "spikes" (Berry et al, 1964) or "club-like" projections (Becker et al, 1967). Such structures were less densely distributed and differently shaped than those of the myxoviruses.…”

Coronaviruses are large, enveloped RNA viruses of both medical and veterinary importance. Interest in this viral family has intensified in the past few years as a result of the identification of a newly emerged coronavirus as the causative agent of severe acute respiratory syndrome (SARS). At the molecular level, coronaviruses employ a variety of unusual strategies to accomplish a complex program of gene expression. Coronavirus replication entails ribosome frameshifting during genome translation, the synthesis of both genomic and multiple subgenomic RNA species, and the assembly of progeny virions by a pathway that is unique among enveloped RNA viruses. Progress in the investigation of these processes has been enhanced by the development of reverse genetic systems, an advance that was heretofore obstructed by the enormous size of the coronavirus genome. This review summarizes both classical and contemporary discoveries in the study of the molecular biology of these infectious agents, with particular emphasis on the nature and recognition of viral receptors, viral RNA synthesis, and the molecular interactions governing virion assembly.

Coronavirus-like serum particles, Australia antigen, and Dane particles were treated by various chemicals and enzymes. Results were studied by electron microscopy. Coronavirus-like particles have no nucleocapsid, no detectable nucleic acid, no antigenic relationships with Australia antigen, and seem to be phospholipoproteins. Similar particles have been found in sera of guinea pigs and in human lipoproteins, especially in the HDL and VHDL fractions. Australia antigen which appears devoid of nucleic acid, and the envelope of Dane particles are sensitive to most agents and behave as phospholipoproteins. Phospholipase C produces ring-like structures. Results differ markedly from those obtained by other workers. The capsid of Dane particles is resistant to most agents. It is separated from the envelope by Tween 80, urea, and other agents. Dane particles seem to contain nucleic acid.
