THE [14C]DEOXYGLUCOSE METHOD FOR THE MEASUREMENT OF LOCAL CEREBRAL GLUCOSE UTILIZATION: THEORY, PROCEDURE, AND NORMAL VALUES IN THE CONSCIOUS AND ANESTHETIZED ALBINO RAT1

Abstract: A method has been developed for the simultaneous measurement of the rates of glucose consumption in the various structural and functional components of the brain in vivo. The method can be applied to most laboratory animals in the conscious state. It is based on the use of z -d e o x y -~ ['4C]glucose (['4C]DG) as a tracer for the exchange of glucose between plasma and brain and its phosphorylation by hexokinase in the tissues. [14C]DG is used because the label in its product, [ '4C]deoxyglucose-6-phosphate, i… Show more

“…The LiCl-induced elevations in k* for AA in central visual and auditory areas may reflect potentiation in these areas of their normal high activity (Mazziotta et al, 1982;Phelps et al, 1981;Sokoloff et al, 1977), which depends on serotonergic, cholinergic, and glutamatergic neurotransmission (Chalmers and McCulloch, 1991;Dringenberg et al, 2003;Ingham et al, 1998). They also may reflect lithium's stimulation of retinal and cochlear inputs to these areas (Jung and Reme, 1994;Pfeilschifter et al, 1988).…”

“…Moreover, tg-6 mice were hemodiluted by an isovolemic blood exchange at a rate of 0.1 ml min À1 with albumin (Human Albumin 5%, Behring Pharmacia, Erlangen, Germany) to a normal target hematocrit of 0.45 (forming group tg-6-HD). In each group, 16 animals were randomized to be used either for the autoradiographic determination of local CBF (n = 8) or for the measurement of local CMR glc (n = 8), according to the methods described in the rat (Sakurada et al, 1978;Sokoloff et al, 1977). These autoradiographic methods were adapted to mice according to the previous use in this species (Jay et al, 1985(Jay et al, , 1988.…”

“…The rat was loosely wrapped with its upper body free in a fast-setting plaster cast that was taped to a wooden block. It was allowed to recover from anaesthesia in a sound-dampened temperature-controlled box for 3 to 4 h, to prevent any effect of anaesthesia on brain metabolism (Kimes et al, 1985;Sokoloff et al, 1977). Arterial blood pressure, heart rate and rectal temperature were measured before and 55 min after injection of saline, D-amphetamine, or raclopride.…”
