The type 1 receptor (CD120a) is the high-affinity receptor for soluble tumor necrosis factor

Abstract: Tumor necrosis factor (TNF) can induce a variety of cellular responses at low picomolar concentrations. This is in apparent conf lict with the published dissociation constants for TNF binding to TNF receptors in the order of 100-500 pM. To elucidate the mechanisms underlying the outstanding cellular sensitivity to TNF, we determined the binding characteristics of TNF to both human TNF receptors at 37°C. Calculation of the dissociation constant (K d ) from the association and dissociation rate constants determi… Show more

“…The observed K D values for the interaction of CD95 with the various CD95L variants were rather high compared with the related ligand-receptor pair TNF-TNFR1 for which K D values between 20 -200 pM have been reported (55)(56)(57)(58). The lower K D value of the CD95L-CD95 interaction is mainly due to slower CD95L association, which takes 8 -12 min to reach 50% of equilibrium binding at concentrations between 5 and 250 ng/ml (Fig.…”

“…Similarly, NO release induced by in vivo administration of soluble TNF was mediated by TNFR1. 8 However, in contrast to soluble TNF, which preferentially activates TNFR1 but not TNFR2, 39 con A exerts its hepatotoxic effects also by expression of transmembrane TNF, which activates both TNFR1 and TNFR2. 7 Hence, TNFR1Ϫ/Ϫ as well as TNFR2Ϫ/Ϫ mice do not release transaminases on con A challenge.…”

“…There are two types of TNF receptors: TNF receptor 1 (TNFR1, p55) is broadly expressed and can be activated by either soluble or transmembrane TNF. TNF receptor 2 (TNFR2, p75) is preferentially activated by the transmembrane form of TNF [122,123] and is expressed by cells of the immune system including microglia but also by macroglial cells. Oligodendrocytes express TNFR2 in a constitutive manner and are induced to express TNFR1 by inflammatory stimuli [124].…”
