The β-barrel membrane protein insertase machinery from Gram-negative bacteria

Abstract: The outer membranes (OM) of Gram-negative bacteria contain a host of β-barrel outer membrane proteins (OMPs) which serve many functions for cell survival and virulence. The biogenesis of these OMPs is mediated by the β-barrel assembly machinery (BAM) complex which is composed of five components including the essential core component called BamA that mediates the insertase function within the OM. The crystal structure of BamA has recently been reported from three different species, including a full-length struc… Show more

“…The overall structure was found in a bent L-shaped conformation, closely resembling that of POTRA3-5 of E. coli BamA (26,39,40). All three POTRA domains contained the conserved βααββ fold observed in other POTRA domains with relatively good structural alignment (23,47,48). Interestingly, the previously observed ∼40-residue insertion within Toc75 POTRA2 (17) overlaps with an extended α-helix encompassing residues 275-296, which we refer to as the P2-helix.…”

“…Given the conservation of key features, these structures serve as models for Toc75, albeit with low sequence identity (7%, 7%, and 8%, respectively). Whereas TamA and BamA have roles in the biogenesis of β-barrel outer membrane proteins in Gram-negative bacteria (23,(52)(53)(54)(55), FhaC has a role in the secretion of filamentous hemagglutinin (FHA) out of Bordetella pertussis (56). Here FhaC serves as the translocon within the outer membrane, first interacting with FHA via its TPS domain and then translocating FHA through its barrel domain, across the outer membrane, and into the extracellular milieu.…”

“…In Escherichia coli, BamA is the central component of the b-barrel assembly machinery (BAM) complex that performs membrane insertion of OM proteins (OMPs) (7). It consists of the characteristic C-terminal b-barrel and five POTRA domains at the N-terminus.…”
