(1) Background: doxorubicin is a potent chemotherapeutic agent, but it has limitations regarding its side effects and therapy resistance. Hydrogels potentially deal with these problems, but several characterizations need to be optimized to better understand how hydrogel assisted chemotherapy works. Poloxamer 407 (P407) hydrogels were mixed with doxorubicin and physico-chemical, biological, and pharmacological characterizations were considered. (2) Methods: hydrogels were prepared by mixing P407 in PBS at 4 °C. Doxorubicin was added upon solutions became clear. Time-to-gelation, hydrogel morphology, and micelles were studied first. The effects of P407-doxorubicin were evaluated on MC-38 colon cancer cells. Furthermore, doxorubicin release was assessed and contrasted with non-invasive in vivo whole body fluorescence imaging. (3) Results: 25% P407 had favorable gelation properties with pore sizes of 30–180 µm. P407 micelles were approximately 5 nm in size. Doxorubicin was fully released in vitro from 25% P407 hydrogel within 120 h. Furthermore, P407 micelles strongly enhanced the anti-neoplastic effects of doxorubicin on MC-38 cells. In vivo fluorescence imaging revealed that hydrogels retained fluorescence signals at the injection site for 168 h. (4) Conclusions: non-invasive imaging showed how P407 gels retained drug at the injection site. Doxorubicin P407 micelles strongly enhanced the anti-tumor effects.

The in situ hydrogel drug delivery system is a hot research topic in recent years. Combining both properties of hydrogel and solution, in situ hydrogels can provide many advantages for drug delivery application, including easy application, high local drug concentration, prolonged drug retention time, reduced drug dose in vivo, good biocompatibility and improved patient compliance, thus has potential in tumor treatment. In this paper, the related literature reports in recent years were reviewed to summarize and discuss the research progress and development prospects in the application of in situ hydrogels in tumor treatment. Abstract.Research Objective: This study aims at observing the clinical effect of the application of the lymphatic targeting drug delivery system in the treatment of malignant tumors. Research Methods: The patients with advanced gastric cancer in our hospital are taken as the samples. According to different treatment methods, they were randomly divided into two groups, namely the observation group and the control group. The patients in the control group were treated with fluorouracil, while the patients in the observation group were treated with compound fluorouracil liposome through the application of the lymphatic targeting drug delivery system in the treatment process.Research Results: In the observation group, a total of six patients experienced adverse reactions, including five patients with nausea and vomiting and one patient with leukopenia, and the incidence rate of adverse reactions was 18.75%. The patients' five-year survival rate was 75%, and the quality of life was 86.32 ± 2.09 points. Compared with the control group, the data differences were statistically significant (p<0.05). Research Conclusions: The application of the lymphatic targeting drug compound fluorouracil liposome in the treatment of gastric cancer is of great clinical value for increasing the patients' disease control rate, improving the patients' living quality and prolonging the patients' life.
