Thidiazuron promotes high frequency regeneration of peanut (Arachis hypogaea) plants in vitro

Abstract: Multiple shoots were induced on Valenciatype peanut (Arachis hypogaea L.) explants cultured in vitro on a nutrient medium supplemented with thidiazuron. Zygotic embryos excised from mature seeds were germinated on Murashige-Skoog nutrient medium, and the resulting plantlets (8 days-old) were used as a source of explants. When cultured on a nutrient medium with increasing levels of thidiazuron (0.5 to 30 mg/l), expiants from various parts of the peanut plant (except the root) produced multiple shoot primordia w… Show more

“…The stimulatory effect of TDZ on multiple shoot formation has been reported earlier by Huetteman and Preece (1993) in woody trees, Kanyand et al (1994) in Arachis hypogea, Ahmad and Anis (2007) in Cyamopsis tetragonoloba and Siddique and Anis (2007) in Cassia angustifolia. Similarly, Ismail et al (2011) successfully regenerated multiple shoots in nodal segments of C. ternatea using TDZ.…”

“…TDZ has been successfully used in plant regeneration systems for many plant species (Faisal et al 2005;Ahmad et al 2006;Ahmad and Anis 2007a) including various legumes (Malik and Saxena 1992;Kanyand et al 1994;Sanago et al 1996;Eapen et al 1998;Victor et al 1999;Ahmad and Anis 2007b;Barik et al 2007). …”

“…This is similar to the results observed in peanut (Kanyand et al, 1994). In a subsequent experiment, a regeneration medium containing 1 mg l Ϫ1 of TDZ was compared with media containing 1 mg l Ϫ1 2,4-D and 1 mg l Ϫ1 BAP (Jähne et al, 1991) or 1 mg l Ϫ1 IAA and 0.2 mg l Ϫ1 kinetin (Wan and Lemaux, 1994) which are often used for barley regeneration.…”

“…Similar results were obtained with TDZ compared to 2,4-D, KIN, and NAA in Sugarcane (Saccharum spp.) (Gallo-Meagher et al 2000), beans (Phaseolus vulgaris) (Malik and Saxena 1992a), chickpea (Cicer arietinum) (George and Eapen 1997), and peanut (Arachis hypogaea) (Kanyand et al 1994). In pigeon pea, higher regeneration of shoot buds (14.7) from the embryonic axes explants was observed on 0.5 mg L −1 TDZ.…”
