Thiols: Role in Oxidative Stress-Related Disorders

Abstract: The effects of oxidative stress occur as a result of peroxidative damage of the macromolecule and membranes of the cells and with the disruption of metabolic activities in the components of the cells in living organisms. Organ and tissue pathologies are known to occur when oxidative stress is excessive in the body. It is known that thiols are one of the main protective mechanisms of the body against oxidative stress. Thiols have been shown to play important roles in enzymatic reactions, apoptosis, detoxificati… Show more

“…Antioxidants are defined as substances that, when present in low concentrations, significantly inhibit or delay oxidative processes while oxidizing themselves, usually in relation to oxidizable substrates. They neutralize free radicals and oxidize themselves by accepting highly reactive unpaired electrons [10,[27][28][29]. It has been indicated by experimental evidence that varied antioxidants play an important role against oxidants in many studies [30][31][32][33][34][35][36][37][38].…”

“…For example, nitric oxide (NO•), which is an RNS, is produced from 1-arginine by nitric oxide synthase and then reacts with superoxide (O2•-) to form peroxynitrite (ONOO-) [9-12]. These reactive oxygen/nitrogen species (RONS), that is, hydroxyl, superoxide anion, nitric oxide, and nitrosyl anion molecules, are highly reactive due to their unpaired valence electrons [10]. Normally, RONS have involved in cell signaling (redox signaling) pathways, thiol switches, regulation of inflammatory cytokines, growth factors, etc.…”

“…Moreover, their metabolic activities in cell components are impaired. Known to tissue and organ pathologies occur in the presence of oxidative stress in the organism [27][28][29][30][31][32][33][34].…”

“…Total thiols-represented by both intracellular and extracellular thiols, as well as thiols bound to proteins-are part of the protective system to counteract oxidative injury. Changes in their status have been shown to coincide with the pathogenesis of stress-associated disorders [22]. To this purpose, the content of free and protein thiols was assessed via Ellman's method after different times of incubation (12,24, and 48 h) of cells in the presence of MPE.…”
