Thrombin Based Photothermal‐Responsive Nanoplatform for Tumor‐Specific Embolization Therapy

Abstract: The specific coagulation in the tumor vasculature has the potential for the ablation of solid tumors by cutting off the blood supply. However, the safe delivery of effective vessel occluding agents in the tumor‐specific embolization therapy remains challenging. Herein, it is reported that the photothermal responsive tumor‐specific embolization therapy based on thrombin (Thr) is delivered by intravenous injection via the phase‐change materials (PCM)‐based nanoparticles. The wax sealing profile of PCM enables sa… Show more

“…Especially for high activity drugs, it is hazardous for preleakage prior to target site even at trace amounts level. Yin et al synthesized a phase-change material (PCM) loaded with both thrombin (Thr) and photothermal reagents IR780 in tumor embolization therapy [ 37 ]. Thr rapidly forms clots in direct contact with blood because of strong coagulation, and this nonspecific embolism is extremely detrimental to normal tissues.…”

“…Owing to the flexibility of polymers and the luminescence from fluorescent nanomaterials (e.g., carbon nanodots and MOFs), nanopolyhybrids can be employed in biosensing, bioimaging, and therapy tools. [172][173][174][175][176] For instance, a glucose-imprinted poly(N-isopropyl acrylamide-acrylamide-vinyl phenyl boronic acid)/nanosized carbon dot hybrid can reversibly change itself depending on the glu-cose concentration and convert biochemical signals to optical signals. The imprinted hybrid composite advanced glucose detection by improving the sensitivity and selectivity over a clinically relevant range of 0-30 mm at physiological pH, which is essential for diabetic monitoring.…”

“…Therefore, PCMs show great potential for utilization as Th carriers. The Dong group developed PCM-based nanocarriers for the safe delivery and controllable release of thrombin (Thr) by coencapsulating IR780 and Thr within PCM via a resolidification approach [ 45 ] (Fig. 6 a).…”
