Thrombospondin 1 is expressed by proliferating mesangial cells and is up-regulated by PDGF and bFGF in vivo

Abstract: Thrombospondin 1 has been shown to be linked to PDGF-mediated mesangial cell proliferation and migration in vitro, but little is known regarding its expression or regulation in glomerular disease. Experimental mesangial proliferative nephritis was induced in rats by injection of anti-Thy1 antibody. Mesangial cell proliferation was associated with de novo expression of thrombospondin 1 mRNA (detected by Northern blot and in situ hybridization) and protein (by Western blot and immunostaining). Although some thro… Show more

“…Fibronectin is present basally within the mesangial matrix functioning in supporting mesangial cell viability, 35 increases in fibronectin have been demonstrated in fibrotic disease processes. 36 TSP is transiently expressed in matrix during development and repair 23,37 and has previously been shown to respond to PDGF, TGF-␤ 1 , and basic fibroblast growth factor, 37 it is a known regulator of TGF-␤ 1 activity. 23,38 We further verify that levels of decorin, a small leucine-rich proteoglycan that acts as an endogenous inhibitor of TGF-␤ 1, 39 are decreased in response to PDGF and are recovered by LXA 4 pretreatment.…”

“…Glomerular preparations were done by the standard sieving procedure as described previously. 25 Biopsies from healthy human and mouse kidneys Normal human renal tissue (n ¼ 6) was obtained from kidneys surgically excised because of the presence of a localized neoplasm. Tissues utilized for this study were obtained from macroscopically normal portions of kidney located at some distance from the neoplastic process.…”

“…25 As secondary antibody, we used a rat adsorbed biotinylated horse anti-mouse IgG (Vector, Burlingame, USA), which was detected by horseradish peroxidase-conjugated avidin D (Vector). The peroxidase substrate 3,3 0 -diaminobenzidine (substrate kit; Vector) was used without nickel resulting in a brown staining (nestin) and Histogreen (Vector) was used for green nuclear staining (PCNA).…”

“…Immunoperoxidase staining for ␣-smooth muscle actin was performed with a murine IgG 2 monoclonal antibody (21) (Sigma) essentially as described previously (22,23). The primary antibody was incubated overnight at 4°C, followed sequentially by biotinylated rabbit antimouse IgG (Zymed Laboratories Inc.), avidin D peroxidase, and finally diaminobenzidine without nickel chloride.…”
