Summary. cc-and @-thyrocalcitonin isolated from pig thyroids were found to be single-chain polypeptides with 32 amino acid residues containing a disulphide bridge. Tryptic digestion yielded inactive hepta-, undeca-and tetradecapeptides, the amino acid composition of which is given. @-thyrocalcitonin proved to be the methionine sulphoxide derivative of the easily oxidizable methionine peptide a-thyrocalcitonin; thus, the designation can be eliminatcd. Both peptides have a potency of about 200 MRC unitslmg dry weight. They can readily bc differentiated by their electrophoretic and chromatographic properties, as well as by their tryptic undecapeptidc, whereas the hepta-and tetradecapeptides, respectively, produced by trypsin are identical.COPP et al. (23. XII. 67) .Summavy. Hog thyrocalcitonin has been isolated in the form of an homogeneous pcptide. After digestion with trypsin three fragments have been separated by paper electrophoresis. The amino acid composition of intact thyrocalcitonin and of the three tryptic fragments is given in a 200 g Acetontrockenpulver werden bci +4" mit 1,2 1 eiskaltem, frisch dcstilliertcin Accton wahrend 15 Min. durchgeruhrt. Nach Absaugcn wird mit 1 1 eiskaltcm, frisch destilliertem Chloroform und dann nochmals mit 1 1 Aceton, w-ie oben, behandelt. Das noch feuchte Pulver wird bei + 4" unter hochtourigem Ruhren rnit cinem Handmixer auf einmal in 2 1 eiskalte Losung, bestchend aus 960 g Harnstoff, 35,l g Cystein-hydrochlorid, 16,6 ml 37-proz. Salzsaure und Wasser ad 2 1, eingetragen und die Suspension so lange gcruhrt bis ein gleichmassig gelartiger Brei entsteht. Dieser wird nun auf einmal in 4 1 eiskalte Aceton-Eisessig-Mischung 1 : 1 eingetragen, wobei sich, nach massigem Riihren, der grosscre Teil auflost. Nach 30 Min. Stehen bei +4" wcrdcn unter intcnsivem Ruhrcn im Lauf von 3/4 Std. nacheinandcr 6 1 eiskaltes Accton und 56 ml 1~ Natriuinchloridlosung zugegeben. Nach dem Abfiltrieren (das Ungeloste wird verworfen) wird zum klarcn 1) Wir danken der Firma SANABO, Wien, fur die Herstellung dieses Praparates.
