TIMP-1 Expression in Human Colorectal Cancer Is Associated with SMAD3 Gene Expression Levels: A Pilot Study

Abstract: The prognosis of colorectal cancer (CRC) varies considerably, and there is a compelling need to identify novel biomarkers with prognostic significance. The aim of the present study was to evaluate the prognostic value of a panel of six genes (CDH1, SMAD3, TGFβ1, ICAM-1, TIMP-1 and MUC12) in CRC patients. METHODS. We evaluated these genes by qRT-PCR in normal and CRC tumor tissue, and correlated the relative gene expression values with clinical, pathological aspects and other biological factors. RESULTS. RNA ex… Show more

“…These adhesion molecules are retrieved in the circulatory system and not only at tumor sites, especially in advanced stages [ 49 , 50 , 51 ]. These adhesion effectors and angiogenic markers could thus be used as biomarkers of invasion and metastasis [ 49 , 50 , 52 , 53 , 54 , 55 ]. Immune and inflammation-related genes may provide a better understanding of the mechanisms through which tobacco smoking causes disease [ 56 ], as well as the possible benefits of immune agonist therapy.…”

“…The coalescent response of integrin–ECM–GF interactions inside the tumor microenvironment promotes the activation and localization of RAS to the inner membrane. Moreover, ECM regulates intercellular communication, cell junction plasticity, and cell adhesion molecules that interact with a wide range of cytokines/chemokine or growth factors [5,69,72,73]. Among the most studied integrin-mediated signaling effectors in cancer cells are the non-receptor tyrosine kinases focal adhesion kinase (FAK) and Src integrins that are linked to downstream signaling effectors such as the Rac1 GTPase and MAPK [74].…”

“…Evidence suggests that MUC expression is involved in the invasion and metastasis of various malignancies, including gallbladder cancer [19], breast cancer [20], ovarian cancer [21], gastric carcinoma [22,23], pancreatic carcinoma [24][25][26], ampullary cancer [27,28], lung cancer [16,29], prostate cancer [30], renal cell carcinoma [31], and appendiceal carcinoma [32]. However, the prognostic value of MUC expression in CRC remains controversial [33][34][35][36][37]. To clarify the inconsistent findings from previously published studies investigating the role of MUC in CRC, this meta-analysis was conducted to assess the association between MUC expression levels and prognosis in CRC and investigate the associations between MUC expression levels and several CRC clinicopathological characteristics.…”
