Tissue metabolic changes for effects of pirfenidone in rats of acute paraquat poisoning by GC-MS

Abstract: We developed a metabolomic method to evaluate the effect of pirfenidone on rats with acute paraquat (PQ) poisoning, through the analysis of various tissues (lung, liver, kidney, and heart), by gas chromatography-mass spectrometry (GC-MS). Thirty-eight rats were randomly divided into a control group, an acute PQ (20 mg kg) poisoning group, a pirfenidone (20 mg kg) treatment group, and a pirfenidone (40 mg kg) treatment group. Partial least squares-discriminate analysis (PLS-DA) revealed metabolic alterations in… Show more

“…IPF has some similarities with the acute effects of SO 2 -induced ALI, both appear to be driven by a mechanism that promote fibroblast recruitment and proliferation leading to a diffuse parenchymal lung disease, resulting in loss of lung function [15,34]. PFD-treatment has shown to have protective effects in other experimental lung disease-models induced by e.g., paraquat, bleomycin and allergen-induced asthma [30,[37][38][39]. In addition to reducing pulmonary fibrosis, PFD-treatment has also been shown to attenuate fibrosis in the liver, heart and kidney [34].…”

“…Currently, metabolomics is generally applied to fields such as pharmacology and toxicology, disease pathogenesis, clinical diagnosis, and plant growth (Bao et al, ; Chen et al, ,b; Jiang et al, ; Ma et al, ). In particular, targeted metabolomics based on triple quadrupole MS has the capacity to perform a robust and precise metabolome analysis owing to its excellent quantitation ability (Chen et al, ).…”
