TMEM30A is essential for hair cell polarity maintenance in postnatal mouse cochlea

Abstract: Background Phosphatidylserine is translocated to the inner leaflet of the phospholipid bilayer membrane by the flippase function of type IV P-tape ATPase (P4-ATPase), which is critical to maintain cellular stability and homeostasis. Transmembrane protein 30A (TMEM30A) is the β-subunit of P4-ATPase. Loss of P4-ATPase function causes sensorineural hearing loss and visual dysfunction in human. However, the function of TMEM30A in the auditory system is unclear. Method… Show more

“…This gene produces an ATPase protein responsible for transferring, or flipping, phosphatidylserine (PS) and phosphatidylethanolamine from the ectoplasmic to the cytoplasmic layers of the cell membrane lipid bilayer. The nature of this function results in the asymmetric partitioning of lipids across the membrane, which is vital for vesicle trafficking, cell signaling, apoptosis, cell survival, and cholesterol/bile homeostasis [49]. The ATP8A2 gene is also expressed within the uterus, where it plays a role in lipid metabolism and cholesterol abundance.…”

“…ABR thresholds were measured at frequencies of 8, 16 and 32 kHz based on visible and reproducible wave II. Detailed methods have been previously described ( Xing et al, 2023 ). The auditory threshold was measured 4 days before and 14 days after noise exposure, respectively.…”
