To get what we aim for – progress in diversity generation methods

Abstract: Protein re‐engineering by directed evolution has become a standard approach for tailoring enzymes in many fields of science and industry. Advances in screening formats and screening systems are fueling progress and enabling novel directed evolution strategies, despite the fact that the quality of mutant libraries can still be improved significantly. Diversity generation strategies in directed enzyme evolution comprise three options: (a) focused mutagenesis (selected residues are randomized); (b) random mutagen… Show more

“…In addition, the new protein sequence of this enzyme will be of great value in the continued efforts to develop protease activity improvements [1820]. One of the key strategies of enzyme engineering is directed evolution through DNA shuffling between different family members [21, 22]. The availability of more unique amino acid sequences from proteases whose activities have been experimentally validated is critical to such projects.…”

“…However, site-saturating mutagenesis will substitute specific sites against all possible amino acid residues. Hence, the importance of a specific amino acid residue towards the function of an antibody can be elucidated through this focused mutagenesis method [78,86]. This can be applied for stability engineering of antibodies by determining the influence of different amino acids at strategic positions along the antibody structure.…”
