Tonic B‐cell and viral ITAM signaling: context is everything

Abstract: The presence of an immunoreceptor tyrosine-based activation motif (ITAM) makes immunoreceptors different from other signaling receptors, like integrins, G-coupled protein receptors, chemokine receptors, and growth factor receptors. This unique motif has the canonical sequence D/Ex(0-2)YxxL/Ix(6-8)YxxL/I, where x represents any amino acid and is present at least once in all immunoreceptor complexes. Immunoreceptors can promote survival, activation, and differentiation by transducing signals through these highly… Show more

“…The B cell antigen receptor also signals independently of ligand engagement and this provides functionally relevant signals in immature B lymphocytes (49) and in diffuse large B-cell lymphoma (50). Significantly, this constitutive signaling is mediated through the B cell receptor ITAMs (51,52). Thus, constitutive signaling appears to be a common feature of YXXL-containing receptors.…”

“…The expression of the BCR is essential for the survival of B cells at all developmental stages, leading to the hypothesis that the BCR must provide constitutive low-level ‘tonic’ signals that promote survival, either spontaneously or on interaction with ligands in the environment 8,9 . Recent genetic and biochemical analyses have shed light on the nature of the tonic signalling pathway, suggesting that the initiation of tonic signalling may not be identical to that of antigen-induced signalling, and may bypass the requirement for the phosphorylation of BCR ITAMs.…”

“…The phosphorylation of the two tyrosines within the ITAM leads to the physical recruitment of Syk to the site of the clustered receptor in an interaction mediated by its tandem pair of SH2 domains [37,38,39]. Interestingly, a number of virally encoded proteins also contain ITAM sequences, which they use to co-opt Syk for their own nefarious purposes [35,40,41]. …”
