Towards the development of a rapid, portable, surface enhanced Raman spectroscopy based cleaning verification system for the drug nelarabine

Abstract: This work shows the potential application of SERS for cleaning verification analysis. SERS may not replace HPLC as the definitive analytical technique, but it could be used in conjunction with HPLC so that swabbing is only carried out once the portable SERS equipment has demonstrated that the manufacturing equipment is below the threshold contamination level.

“…Although NIR is gaining most of the popularity in food and pharmaceutical fields, the absorption coefficients of the vibrational modes that are observed in the NIR are weaker than those associated with mid‐IR vibrations leading to weaker spectral intensities with broad and overlapping bands (Druy 2004). Surface enhanced Raman spectroscopy was reported for analysis of nelarabine providing the required levels of sensitivity for cleaning validation (Corrigan and others 2010). Nevertheless, it was suggested as a semi‐quantitative limits test due to poor replicate precision related to the heterogeneous drying pattern of nelarabine on the SERS active surface (Corrigan and others 2010).…”

Infrared spectroscopy has been recognized as a sensitive optical absorption technique for detecting low concentrations of organic compounds on reflective surfaces such as metal (Griffiths and deHaseth 1986; Mehta and others 2003).…”
