Transcript expression of vesicular glutamate transporters in lumbar dorsal root ganglia and the spinal cord of mice – Effects of peripheral axotomy or hindpaw inflammation

Abstract: Using specific riboprobes, we characterized the expression of VGLUT1-VGLUT3 transcripts in lumbar 4-5 (L4-5) DRGs and the thoracolumbar to lumbosacral spinal cord in male BALB/C mice after a 1- or 3-day hindpaw inflammation, or a 7-day sciatic nerve axotomy. Sham animals were also included. In sham and contralateral L4-5 DRGs of injured mice, VGLUT1-, VGLUT2- and VGLUT3 mRNAs were expressed in ~45%, ~69% or ~17% of neuron profiles (NPs), respectively. VGLUT1 was expressed in large and medium-sized NPs, VGLUT2 … Show more

“…These include the vesicular glutamate transporter 2 (VGLUT2), the vesicular glutamate transporter 3 (VGLUT3), and EAAT3/EAAC1 [51; 64; 70]. Since both VGLUT2 and EAAT3/EAAC1 expression is associated with nociceptive transmission [20; 63], we used Western blot to assess for possible change in protein levels.…”

“…PKC-γ is expressed in excitatory interneurons in the deep part of laminae II and III (35). Glutamatergic VGLUT2 + neurons are abundant throughout the spinal cord (3,36). To identify VGLUT2 + nuclei, we used a BAC-VGLUT2::Cre transgenic mouse line (31) (which Phenotyping of Dorsal Horn Neurons: NECAB1.…”

“…In the current study, a large population of rat ICG neurons contained GLS indicating that many ICG neurons produce glutamate. Glutamatergic neurons use vesicular glutamate transporters (VGLUTs) 1–3 for packaging neurotransmitter glutamate into synaptic vesicles and VGLUTs are considered reliable markers for glutamatergic neurons (Takamori et al, 2000, 2001, Malet et al, 2013). Among them, VGLUT1 and VGLUT2 have been more intensively studied due to the availability of reliable specific antibodies.…”
