Transcription profile analysis of the endometrium revealed molecular markers of the personalized ‘window of implantation’ during in vitro fertilization

Abstract: To determine the most informative markers for assessing the functional state of endometrium during the 'window of implantation' and creating a model for assessment of the readiness of endometrium for embryo implantation. Forty-seven women with tubal infertility and a successful IVF pregnancy participated in the study. Pipelle endometrial sample was performed during the supposed 'window of implantation' in natural cycle with subsequent histological study, and transcriptional profile of genes GPX3, PAEP, DPP4, T… Show more

“…The PAEP gene showed the greatest upregulation in the mid-secretory endometrium in this study. This gene has been reported to display negative or low expression in the proliferative endometrium (32) and may serve as a candidate biomarker for endometrial receptivity (33,34). SFRP4 modulates Wnt signaling and is more highly expressed in proliferative endometrium compared with that in secretory endometrium (35); upregulation of SFRP4 in the placenta is associated with severe preeclampsia (36).…”

“…The 24 transcripts contained several genes that have previously been associated with genital development, sex hormone actions, and/or male reproduction (Table 1) [ [21][22][23][24][25][26][27][28]. In particular, APOD is known as an androgentarget gene in human genital fibroblasts, and Cyp1b1 has been implicated in genital development of mice.…”

“…A total of 6,366 DEGs were similarly upregulated or downregulated in primary EMSFs and D14 EBs compared with hiPSCs (adjusted p < 0.05), as demonstrated by the Venn diagram in Figure 5 D. Thirty genes (15 upregulated and 15 downregulated genes in D14 and primary EMSFs compared with hiPSCs) with the highest fold changes are listed in the clustered heatmap ( Figure S5 A). The top 15 downregulated DEGs include pluripotency markers such as SOX2 and NANOG , whereas the top 15 upregulated genes include genes that play crucial roles in endometrial function, such as WNT2 , GBP1 , and POSTN ( Burmenskaya et al., 2017 , Kumar et al., 2001 , Li et al., 2017 ). Endometrium-specific transcription factors ( HOXA9 , HOXA10 , HOXA11 , and PGR ) and EMSF markers ( MME , COL1A1 , FN1 , and VIM ) were present among other common DEGs.…”
