Transgenerational Effects on Calf Spermatogenesis and Metabolome Associated with Paternal Exposure to the Fukushima Nuclear Power Plant Accident

Abstract: The Fukushima nuclear power plant (FNPP) accident raised worldwide attention to the health risk of radiation exposure and to its potential transgenerational effects. Here, we analysed transgenerational effects on calf spermatogenesis and on blood plasma metabolome in order to detect alterations associated with paternal exposure to low-dose-rate (LDR) radiation. Sperm was collected from a bull exposed to radiation for 2 years abandoned in the ex-evacuation zone of the FNPP accident (the abandoned bull) and was … Show more

“…The acrosomal integrity of seven raccoon sperm samples was assessed by fluorescein isothiocyanate‐conjugated peanut agglutinin (FITC‐PNA) staining (Sigma‐Aldrich) as described previously (Suzuki et al, 2020) and categorised as follows: (a) intense acrosomal cap fluorescence indicating an intact acrosome; (b) acrosomal cap fluorescence indicating a partially damaged acrosome; and (c) no fluorescence indicating complete outer acrosomal membrane loss.…”

“…Following the Fukushima Daiichi Nuclear Power Plant (FDNPP) accident, large amounts of radioactive substances, particularly volatile elements, such as radioactive iodine, caesium and tellurium, were released into the environment. The ex‐evacuation zone comprises a 20‐km‐radius area around the FDNPP, from which we have established an archive system of livestock and wild animals (Ariyoshi et al., 2018; Fukuda et al., 2013; Fukuda et al., 2015; Fukuda et al., 2016; Koarai et al., 2016; Koarai et al., 2018; Koarai et al, 2020; Ohdaira et al., 2019; Suzuki et al, 2020; Takahashi et al., 2015; Takino et al., 2017; Urushihara et al., 2016; Meguro et al., 2019; Urushihara et al., 2018; Yamashiro et al., 2013; Yamashiro et al., 2014; Yamashiro et al., 2015).…”

“…Radiation metabolomics has often been employed to diagnostically or therapeutically search for relevant biochemicals after exposure to ionizing radiation [ 46 , 47 , 48 , 49 , 50 ]. Metabolomics was used to investigate metabolite changes in rice seeds [ 15 , 18 ] and calf blood plasma [ 51 ] after the Fukushima nuclear accident. To demonstrate that the creeping wood sorrel responds to the low dose of radiation that is relevant in the field, we used contaminated soil collected from Fukushima as the radiation source.…”
