Treatment of Pimpinella Alpina Molk Improve Oxidative stress and Inhibit Liver Cellular Apoptosis in Rats Following UVB Irradiation: Is there Any Correlation?

Abstract: Objective: The effect of Pimpinella alpina Molk (PaM) in improvement of oxidative stress has been established, but did not for cellular apoptosis. This investigation was conducted to evaluate the effect of PaM on GPx and XO activities, hepatocytes apoptosis and their correlation following UVB radiation. Methods: Forty male rats were assigned into 8 groups: Nor-G, Neg-7, Neg-15, PaM50-7, PaM100-7, PaM150-7, PaM100-15, and PaM150-15. UVB irradiation was given 10 minutes to each rat daily, blood and liver organ s… Show more

“…This result implied that zamzam water extract of CZ treatment by the dose of 100 and 500 µl on T47D cell line was unable to trigger DNA damage largely and subsequently apoptosis. In the biological system, apoptosis is used to delineate the intrinsic cell suicide program, and morphologically is characterized by global cell shrinkage, cell blabbing, and chromatin condensation accompanied by nuclear and DNA fragmentation into specific fragment sizes form DNA ladder pattern 22 . Initiation of apoptosis by the cell is determined by gene activities, leading to cellular self-destruction through either intrinsic or the extrinsic or both pathways 23 .…”

“…The result of the present study has also supported the theory that, apoptosis is a process triggered by death signal induction derives from both internal and external milieu, inducing mitochondrial membrane to release cytochrome-c. Cytochrome-c holoform along with apaf-1, procaspase-9, and Adenosintriphosphat (ATP) to form apoptosomedeath complex, that proteolitically convert procaspase-9 to caspase-9, and then activate caspase-7, caspase-6, caspase-3, and finally induce DNA fragmentation and cell death 25 . On the other hand, apoptosis can be prevented by Bcl-2 (a protein anti apoptosis) through releasing cytochrome-c and diablo/ Smac, an Inhibitor Apoptosis Protein (IAP) binding, originated from mitochondria.…”

“…Pimpinella alpina Molk (PaM) treatment with 100-150 mg daily dose during 7 days have been proven capable of inhibiting liver apoptosis characterized by decrease in mRNA protein expression of Bax and Caspase-3 following ultraviolet B (UVB) light irradiation in rats. 1 Considering cell is able to denovo synthesis protein and other building blocks in order to regenerate their own self structure following damage from variety insults, 2-5 therefore the emerging question is, whether treatment with PaM in the same doses (100-150 mg daily) during 15 days will reduce apoptosis marked by Bax and Caspase-3 protein expression significantly compared to those of 7 days treatment following UVB irradiation. In addition, in this study, identification of Bax and Caspase-3 protein expression in liver cells will be assessed by immunohistochemical (IHC) staining method to confirm the previous result were measured by Rt-PCR method.…”

“…6,7 The most powerful and damaging effect of UVB on skin is mostly attributed to reactive oxygen species (ROS) particularly radical hydroxyl (OH • ). 1 Another ROS which is also produced in skin triggered by UVB are hydrogen peroxide (H 2 O 2 ) and oxygen superoxide (O 2 -) resulted from mitochondrial oxidative phosphorilation. 1,8 Furthermore, H 2 O 2 is rapidly circulated in systemic circulation and may be degraded to OH • through Fenton or Heber Weis reaction following UVB radiation.…”
