Triboelectrification and dissolution property enhancements of solid dispersions

Abstract: The use of solid dispersion techniques to modify physicochemical properties and improve solubility and dissolution rate may result in alteration to electrostatic properties of particles. Particle triboelectrification, plays an important part in powder processing, affecting end product quality due to particle deposition and powder loss. This study investigates the use of glucosamine hydrochloride (GLU) in solid dispersions with indomethacin. Solvents selected for the preparation of the dispersions were acetone,… Show more

“…Triboelectrification has been used to study the impact of the counter ion on flurbiprofen salts as a consideration during the preformulation process [22], reduce the charging of flurbiprofen (a drug with a high propensity for charging) in binary mixtures of cellulose ethers [23], ordered mixing [24] and recently as a way of manipulating the charge of their final product of solid dispersions using single solvents and binary mixtures of solvents [25].…”

“…The pure CBZ, GLU and their co-ground samples were characterised also using FT-IR according to the methodology detailed by Asare-Addo et al [25]. In brief, a 5-10 mg sample of pure CBZ, pure GLU, Physical mix of CBZ and GLU, CBZ and GLU ground for either 10 min or 60 min (all at a 1:1 ratio) were placed on an attenuated total reflection (ATR) plate and analysed using FTIR spectroscopy (Nicolet 380 FT-IR spectrometer, ThermoElectron Corporation, USA).…”

“…The poor solubility of drugs has always been a major problem in pharmaceutical development and this problem is now more prevalent with more than 40 % of the new chemical entities being practically insoluble in water or lipophilic in nature [3][4][5][6][7]. As dissolution rates are typically the rate-limiting step for bioavailability, especially for poorly soluble drugs, enhancement of solubility is vital to attaining suitable systemic concentrations for therapeutic effect [8].…”

“…Several authors have classed the solid dispersion approach as one of the most effective method of improving dissolution of drugs [5,8,22]. It involves the dispersion of one or more active ingredients in an inert excipient or carrier, where the active ingredients could exist in a finely crystalline, solubilised or amorphous state [23,24].…”
