“…Our previous data had shown that 8-OHdG inhibitor NAC mitigated TCSmediated decrease of GDM in HepG2 cells (Ma et al, 2013). In this study we found that 8-OHdG increased in low-dose TCS exposed HepG2 cells, suggesting that the low-dose TCS induced oxidative stress (Tamura et al, 2012), also the methylation levels of LINE-1 gene data supported that 8-OHdG decrease may impair global DNA hypomethylation. In summary, we believe that the increased 8-OHdG levels played an indirect role in inducing genomic hypomethylation in TCS exposed HepG2 cells.…”

“…A recent study shows that TCS can act as H 2 O 2 by decreasing the contents of intracellular thiol, contributing to oxidative stress, which may damage genomic DNA to accumulate 8-OHdG in the genomic DNA (Tamura et al, 2012). Indeed, a previous study has suggested that 8-OHdG can inhibit the formation of 5mdC in CpG dinucleotides (Valinluck et al, 2004).…”

“…Other compounds, such as triclosan (Tamura et al, 2012) and potentially paraquat and irgarol (Walker et al, 2008), can cause an increase in the intracellular concentration of free zinc(II) and illicit gene expression via the MTF1 pathway.…”

“…For example, the promoter region of rainbow trout MT-A possesses six MREs, four of which are arranged in tandem (Olsson et al, 1995), and MT-B possesses four MREs (Samson and Gedamu, 1995). Metals may displace Zn from metal binding sites within the cell, increasing intracellular free zinc and inducing MT expression.Other compounds, such as triclosan (Tamura et al, 2012) and potentially paraquat and irgarol (Walker et al, 2008), can cause an increase in the intracellular concentration of free zinc(II) and illicit gene expression via the MTF1 pathway. …”
