TRPC1 is a differential regulator of hypoxia-mediated events and Akt signalling in PTEN-deficient breast cancer cells

Abstract: Hypoxia is a feature of the tumour microenvironment that promotes invasiveness, resistance to chemotherapeutics and cell survival. Our studies identify the transient receptor potential canonical-1 (TRPC1) ion channel as a key component of responses to hypoxia in breast cancer cells. This regulation includes control of specific epithelial to mesenchymal transition (EMT) events and hypoxia-mediated activation of signalling pathways such as activation of the EGFR, STAT3 and the autophagy marker LC3B, through hypo… Show more

“…Pharmacological inhibition or silencing of some of these calcium channels or pumps reduces breast cancer cell proliferation and/or invasiveness in in vitro and in vivo studies (Azimi, Roberts-Thomson, & Monteith, 2014). Recently the Ca 2+ permeable ion channel TRPC1 was identified as a positive modulator of pAKT levels in PTEN-deficient breast cancer cell lines, whereby TRPC1 silencing decreases basal pAKT levels (Azimi, et al, 2017). The potential role of other calcium channels or even specific calcium pumps to regulate pAKT in PTEN-deficient breast cancer cells has not been fully explored.…”

“…Images were acquired using the Chemidoc Touch Imaging System (Bio-Rad), analyzed using the ImageLab software version 5.2.1 (Bio-Rad) and local background subtraction. Protein density was normalized to beta actin loading controls and pAKT protein expression was normalized to total AKT protein expression as previously described (Azimi, et al, 2017).…”

“…Considering that compressive stress stretches cell membrane and thus increases membrane tension, it may as well alter cancer cells' behaviors through tensionmediated conformational changes of proteins and lipids in the membrane 15 . In particular, the increase of membrane tension can activate several stretch-activated ion channels (SAC) including Piezo and transient receptor potential (TRP) channels [16][17][18] .…”
