Tsc Gene Locus Disruption and Differences in Renal Epithelial Extracellular Vesicles

Abstract: In tuberous sclerosis complex (TSC), Tsc2 mutations are associated with more severe disease manifestations than Tsc1 mutations and the role of extracellular vesicles (EVs) in this context is not yet studied. We report a comparative analysis of EVs derived from isogenic renal cells except for Tsc1 or Tsc2 gene status and hypothesized that in spite of having similar physical characteristics, EVs modulate signaling pathways differently, thus leading to TSC heterogenicity. We used mouse inner medullary collecting … Show more

“…Characterization methods: Additionally, the advancement of imaging technologies, such as transmission electron microscopy (TEM) and scanning electron microscopy (SEM), has allowed for the visualization of EVs at higher resolutions, providing insights into their structure and biogenesis [ 33 ]. The advancements in the field of genomics and proteomics have also played a critical role in EV research, by allowing the characterization of the molecular composition of EVs at a large scale [ 18 , 34 ]. This has led to the identification of new biomarkers and therapeutic targets for various diseases.…”

“…Tsc1 and Tsc2 control cell proliferation independent of each other, and only Tsc2 function is affected by p27 expression [ 75 ]. Further evidence for separate functions of Tsc1 and Tsc2 comes from microarray and proteomic approaches, which reveal that each TSC gene triggers substantially different cellular responses [ 77 , 78 , 79 , 80 , 81 , 82 ].…”
