Tumor and endothelial cells collaborate via transcellular receptor complexes

Abstract: Many cellular signaling pathways are initiated by cell‐surface ligand‐sensing complexes that incorporate not just one but multiple receptors. Most studies focus on receptors coexpressed on a single cell (cis interactions), but complexes containing receptors on adjacent cells (trans interactions) are also possible. Recent work by Morin et al published in this journal provides critical evidence for such trans interactions between Neuropilin‐1 (NRP1) expressed on human tumor cells and vascular endothelial growth … Show more

“…NRP-1 promotes vascular endothelial growth factor receptor2 (VEGFR2) signal transduction induced by galectin-1 [ 36 ]. Meanwhile, VEGFR2/NRP-1 trans-complex reduces angiogenesis and aggravates the hypoxic environment of PDAC [ 37 , 38 ], which promotes the activation of TAM and MDSC. Interestingly, IL-8 secreted by CAFs can promote high expression of NRP-1, while in PDAC, galectin-1 can induce more IL-8 secretion by CAFs [ 39 ].…”

“…The formation of cis NRP1-VEGFA-VEGFR2 complexes within cells plays a crucial role in enhancing angiogenesis. [ 76 – 78 ] However, trans-NRP1-VEGFA-VEGFR2 complexes across cells play an inhibitory role in angiogenesis. [ 78 ] Pan et al [ 79 ] using a mouse xenograft tumor model, determined that antibodies that blocked VEGFA binding to NRP1 enhanced the anti-tumor effect of anti-VEGFA antibodies.…”
