Tumor-selective catalytic nanomedicine by nanocatalyst delivery

Abstract: Tumor cells metabolize in distinct pathways compared with most normal tissue cells. The resulting tumor microenvironment would provide characteristic physiochemical conditions for selective tumor modalities. Here we introduce a concept of sequential catalytic nanomedicine for efficient tumor therapy by designing and delivering biocompatible nanocatalysts into tumor sites. Natural glucose oxidase (GOD, enzyme catalyst) and ultrasmall Fe3O4 nanoparticles (inorganic nanozyme, Fenton reaction catalyst) have been i… Show more

“…Amorphous silica, whose long-range formula is SiO2, consists of tetraoxosilicon centres (denoted Q for quadrifunctional) interlinked by siloxane bonds. 21 A fully condensed Si centre (Q 4 , being linked to 4 other centres) can only exist in the core of a particle, whereas partially condensed centres (Q 2 or Q 3 ) are associated with the surface. USSN accounted for most (84  3%) of the silicon content in the dispersion and, of this, 43% was partially condensed (Q 2 & Q 3 ) and 57% was fully condensed (Q 4 ).…”

“…Thus, cancer cells are more sensitive to further enhanced oxidative stress beyond the cellular tolerability threshold 2 . On this basis, ROS-mediated therapies, such as photodynamic therapy (PDT) [3][4][5][6] and chemodynamic therapy (CDT) [7][8][9][10][11] , are developed to disrupt the cellular self-adaptation mechanisms and induce cell death based on ROS-generating agents 12 .…”

“…[23] Moreover, under the intracellular superoxide dismutase (SOD)mediated disproportionation reactions, O 2 −• molecule further converts to hydrogen peroxide (H 2 O 2 ) and O 2 . [27,28] Remarkably, O 2 is partially recyclable during this cascade reaction and valuable to ameliorate the hypoxia microenvironment for enhanced PDT efficacy.
