Tumor-Specific Immunoenhancing Effects after Local Cryoablation for Metastatic Bone Tumor in a Mouse Model

Abstract: We investigated the abscopal effect after cryoablation (CA) on bone metastasis using a mouse model. Breast cancer cells were implanted in the bilateral tibiae of mice. The left tumor was treated locally with CA, and the right abscopal tumor (AT) was left untreated. The mice were divided into four groups based on the combination of CA and intraperitoneal administration of anti-PD-1 antibody (PD) as treatment interventions (Control, CA, PD, and CA + PD). The reduction ratio of the size of AT, the quantitative im… Show more

“…An additional promising advantage of cryoablation is that it may favour a specific immune response against tumours, which may stimulate or enhance the combined efficacy of antitumour immunotherapy. [27][28][29][30][31] Compared with the percutaneous approach, the transbronchial approach seems to be a promising therapeutic alternative with a lower risk of pneumothorax. 9 32 Previous studies have reported that transbronchial RFA and MWA are feasible for the treatment of early-stage peripheral lung cancer.…”

“…In addition, regulation of the rate of thawing equally can produce various necrotic effects [11]. Other studies proposed that, beside these effects, there would be also a systemic immunomodulatory response produced because of the sistemical dissemination of tumor antigens [12,13].…”

“…Since the 1960s, cryotherapy has been known to have an aberrant immunoregulatory effect known as the "abscopal effect," and some studies have shown that it can affect or potentially eliminate metastatic tumors [10][11][12]. The necrotic cells persisted in the body after cryotherapy, which could trigger a robust immunological response.…”
