Tumour cell-activated platelets modulate the immunological activity of CD4+, CD8+, and NK cells, which is efficiently antagonized by heparin

Abstract: Platelets, key players in haemostasis, are progressively investigated with respect to their role in immunity and inflammation. Although the platelet support to haematogenous cancer cell metastasis has been the subject of multiple studies, their impact on anti-cancer immunity remains unaddressed. Here, we investigated the immunomodulatory potential of platelets upon their activation by MDA-MB-231 breast cancer cells in various in vitro approaches. We provide evidence that platelets as well as their tumour cell-… Show more

“…[330][331][332] In addition, heparin has been reported to be able to counteract the immunosuppressive effects of tumor cellactivated platelets, which were brought on by platelet-mediated Treg cell differentiation, decreased NK cell activity, and inhibited CD8 + T cells. [333] Wei et al created the nano-delivery platform (LT-DOX/LT-IMQ) using self-assembled polymeric micelles (LT) made of LMWH and TOS. In acidic conditions, LT-DOX/LT-IMQ could rapidly release the drugs DOX and IMQ.…”

“…Several publications have demonstrated non-anticoagulation effects of heparin, such as binding to inflammatory factors, neutralization of the complement factor C5a, prevention of neutrophil chemotaxis and migration of leukocytes, and isolation of acute inflammatory proteins. Heparin can interact with many proteins involved in the pro-inflammatory and pro-coagulant cascade to guard against sepsis-associated inflammation and coagulation abnormalities ( Li and Ma, 2017 ; Gockel et al, 2022 ; Lippi et al, 2022 ). In addition, in animal models of chronic myocarditis, heparin was found to be associated with low rates of myocardial inflammation and collagen deposition ( Frizelle et al, 1992 ).…”
