Tumour exosomes display differential mechanical and complement activation properties dependent on malignant state: implications in endothelial leakiness

Abstract: BackgroundExosomes have been implicated in tumour progression and metastatic spread. Little is known of the effect of mechanical and innate immune interactions of malignant cell-derived exosomes on endothelial integrity, which may relate to increased extravasation of circulating tumour cells and, therefore, increased metastatic spread.MethodsExosomes isolated from non-malignant immortalized HCV-29 and isogenic malignant non-metastatic T24 and malignant metastatic FL3 bladder cells were characterized by nanopar… Show more

“…In this study, an in‐depth examination of a broad range of EVs is undertaken, providing insights to the largely unexplored field of mechanics of natural vesicles. Very few previous pioneering studies addressed the importance of mechanical properties of extracellular vesicles and so far the pressurization of vesicles due to surface interactions has not been taken into account. Our recently developed approach does take pressurization into account.…”

“…It has been confirmed that AFM can measure the relative size distribution of exosomes under physiological conditions . Compared with other techniques, AFM can simultaneously measure the size distribution of exosomes and map their mechanical properties with nanometer accuracy . AFM can simultaneously quantify and detect the abundance, structure, biomechanics and other characteristics of exosomes in tumor samples …”

“…Labeled purified exosomes with fluorescent agents could enhance the assessment of targeting accuracy, cellular uptake, internalization manner, cellular location, distribution, and accumulation in cells and body . Due to lipoid essence of exosomes, fluorescent lipophilic dyes such as PKH67, PKH26, rhodamine B (oh R18), Dil, DiD, and DiRwere effectively used.…”

“…158 Labeled purified exosomes with fluorescent agents could enhance the assessment of targeting accuracy, cellular uptake, internalization manner, cellular location, distribution, and accumulation in cells and body. 238 Due to lipoid essence of exosomes, fluorescent lipophilic dyes such as PKH67, PKH26, rhodamine B (oh R18), Dil, DiD, and DiR 158,188,[245][246][247][248][249] were effectively used. Also, the staining via membrane-permeable chemical compounds such as carboxyfluorescein succinimidyl ester and 5(6)-carboxyfluorescein diacetate by entering into exosomes was performed.…”
