Tumour imaging by the detection of fibrin clots in tumour stroma using an anti-fibrin Fab fragment

Abstract: The diagnosis of early and aggressive types of cancer is important for providing effective cancer therapy. Cancer-induced fibrin clots exist only within lesions. Previously, we developed a monoclonal antibody (clone 102-10) that recognizes insoluble fibrin but not fibrinogen or soluble fibrin and confirmed that fibrin clots form continuously in various cancers. Here, we describe the development of a Fab fragment probe of clone 102-10 for tumour imaging. The distribution of 102-10 Fab was investigated in geneti… Show more

“…Immunohistochemistry (IHC) was carried out as previously described. 15 Anti-TMEM180 antibody or anti-EGFR mAb (cetuximab, Merck, Darmstadt, Germany) was labeled with HRP using Peroxidase Labeling Kit-SH (Dojindo). Tissues were then incubated with the HRP-conjugated anti-TMEM180 mAb or anti-EGFR mAb for 1 hour at room temperature.…”

“…Cross‐linked fibrin and FBG‐related proteins are deposited extravascularly from the local hypermeable blood vessels within tumor stroma, and they play important roles in the pathogenesis of tumor growth and spread . The tumor stroma and the local ECM is a versatile environment where coagulation‐related events (i.e., platelet activation, fibrin clot deposition, fibrinolysis) are activated by cancer, and their biochemical composition is dramatically affected by tumor cell growth and its further metastasis, accompanied by tumor‐driven production and release of specific factors (e.g., proinflammatory, prothrombotic, and proangiogenic) …”
