Tunable, Ultrasensitive pH‐Responsive Nanoparticles Targeting Specific Endocytic Organelles in Living Cells

Abstract: Switch it up: Tunable, pH‐responsive nanoparticles can be selectively activated in different endocytic compartments. At high pH values, micelle formation (see picture, left) quenches fluorescence by Förster resonance energy transfer. The micelles disassemble at low pH values, leading to fluorescence emission. This nonlinear on/off nanoplatform offers many exciting opportunities in diagnostic imaging and drug‐delivery applications.

“…Nanoparticles were not able to enter the cell in the media at pH 7.4, whilst at pH between 6.3 and 5.4 were found be enabled in different cellular pH compartments, such as lysosomes or early endosomes [37] which could be used in drug delivery applications.…”

“…Such dependence is highly reversible. As shown in figure 2D, by repeating switching pH from 6 to 10, very little variation in the ratio, which is different from those irreversible pH sensors derived from micelle or avidin loaded with TARMA [11,14] and other known ratiometric pH indicators [15] . The pH response of TG-AuNPs was still independent of the concentration ( Figure S3), indicating the changes in the fluorescence intensity at different values were not induced by the aggregation of AuNPs.…”
