Two Common Mitochondrial DNA Polymorphisms are Highly Associated with Migraine Headache and Cyclic Vomiting Syndrome

Abstract: Mitochondrial dysfunction is a hypothesized component in the multifactorial pathogenesis of migraine without aura (MoA, 'common migraine') and the related condition of cyclic vomiting syndrome (CVS). In this study, the entire mitochondrial genome was sequenced in 20 haplogroup-H CVS patients, a subject group studied because of greater genotypic and phenotypic homogeneity. Sequences were compared against haplogroup-H controls. Polymorphisms of interest were tested in 10 additional CVS subjects and in 112 haplog… Show more

“…The strong association between CVS and maternal migraine [11] has spurred research into the possibility of maternal inheritance of mitochondrial disease. CVS has been associated with several mitochondrial DNA polymorphisms including 19519C-T [12] and variants in the hypervariable segments in the mitochondrial sequence [13].…”

“…The cause is likely a result of disorders of the central neural pathways and neuroendocrine mediators involved in the brain-gut pathways of nausea and vomiting [3]. There is also some evidence to suggest underlying abnormalities in the mitochondria [5][6][7] and the autonomic nervous system [8][9][10][11][12].…”

“…Cyclic vomiting syndrome (CVS) is a functional disorder that generally demonstrates maternal inheritance (2); mtDNA sequence variants have been reported in CVS (3,24,42,44). In a previous study by Boles et al (2), two common mtDNA single nucleotide polymorphisms (SNPs), 16519C (cytosine)ϾT (thymine) and 3010G (guanine)ϾA (adenine), were found to be associated with CVS and migraine among patients with the mtDNA SNP 7028C (haplogroup H).…”
