Two‐dimensional Hydrophilic Interaction/Reversed‐phase Liquid Chromatography for the Preparative Separation of Polar and Non‐polar Taxanes

Abstract: Combination of hydrophilic and reversed-phase chromatography in two-dimensional liquid chromatography (2D-LC) systems is a useful method for the separation of compounds with a large polarity range such as taxanes.

“…Two-dimensional RP-RP LC is a practical separation method and has been used for isolation of compounds from pharmaceutical samples [22]. Two-dimensional HILIC coupled with RP chromatography has been proved to be very useful for separation of the components of Taxus baccata L. [23]. Although these preparative 2D HPLC systems enable reasonable separation of compounds with a wide range of polarity and can, to some extent, solve the problem of peak capacity and separation selectivity, there are still specific deficiencies in comprehensive characterization of weakly polar TCM extracts, because of limitations of the modes of separation and the complexity of the samples.…”

A reasonable chromatographic method based on multidimensional heart‐cutting HILIC and RPLC for simultaneous separation of polar and nonpolar taxanes was introduced .
