Two-faced Janus: the dual role of macrophages in atherosclerotic calcification

Abstract: Calcification is an independent predictor of atherosclerosis-related cardiovascular events. Microcalcification is linked to inflamed, unstable lesions, in comparison to the fibrotic stable plaque phenotype generally associated with advanced calcification. This paradox relates to recognition that calcification presents in a wide spectrum of manifestations that differentially impact plaque’s fate. Macrophages, the main inflammatory cells in atherosclerotic plaque, have a multifaceted role in disease progression.… Show more

“…The role of inflammation and immune cells in the development of various plaque types is incompletely understood. Circulating inflammasome particles may be an important component in this process by increasing monocyte adhesion to endothelial cells ( Waring et al, 2021 ).…”

“…In Trem2 hi macrophages, researchers observed increased levels of cathepsins, which could increase plaque vulnerability and the inflammation of atherosclerosis [ 55 ]. Trem2 may also contribute to the microcalcification of atherosclerotic lesions that can worsen local tissue stress and impair plaque stability [ 56 , 57 ].…”

“…Macrophages, on the other hand, play a key role in the regulation of VSMC/VIC-mediated VC [ 28 ]. Macrophages can differentiate into different subpopulations, such as pro-inflammatory or anti-inflammatory, depending on their microenvironmental signals.…”
