Two new mouse models of Gjb1‐associated Charcot–Marie–Tooth disease type 1X

Abstract: BackgroundCharcot‐Marie‐Tooth disease type 1X is caused by mutations in GJB1, which is the second most common gene associated with inherited peripheral neuropathy. The GJB1 gene encodes connexin 32 (CX32), a gap junction protein expressed in myelinating glial cells. The gene is X‐linked, and the mutations cause a loss of function.AimsA large number of disease‐associated variants have been identified, and many result in mistrafficking and mislocalization of the protein. An existing knockout mouse lacking Gjb1 e… Show more

“…Samples were reduced with 10 mM dithiothreitol at 42 o C for 30 minutes on a ThermoMixer with 500 rpm agitation, alkylated with 15 mM iodoacetamide for 20 minutes at room temperature on a ThermoMixer with 500 rpm agitation, and trypsin-digested (Sequence Grade Modified; Promega) with a 1:50 trypsin:protein ratio at 37 o C for 20h. Following the digest, all peptide samples were C18-purified using Millipore C18 zip-tips (Millipore, Cat#: ZTC18S096) according to the manufacturer protocol's and as previously reported (48). Eluted purified peptides were dried with a vacuum centrifuge and stored at -20 o C until tandem mass tag (TMT) labeling was performed.…”

“…Liquid Chromatography Tandem Mass Spectrometry Analysis (LC-MS/MS): Each of the dried multiplexed peptide samples were reconstituted in 25 uL of LC-MS grade water with 0.1% TFA and zip-tipped using Millipore C18 zip-tips according to the manufacturer protocol's and as previously reported (48). Purified multiplexes were then dried in a vacuum centrifuge and reconstituted in 20 µL of 98% H2O/2% ACN with 0.1% formic acid via pipetting and vortexing.…”
