Two New Polyphenol Oxidase Genes of Tea Plant (Camellia sinensis) Respond Differentially to the Regurgitant of Tea Geometrid, Ectropis obliqua

Abstract: Polyphenol oxidases (PPOs) have been reported to play an important role in protecting plants from attacks by herbivores. Though PPO genes in other plants have been extensively studied, research on PPO genes in the tea plant (Camellia sinensis) is lacking. In particular, which members of the PPO gene family elicit the defense response of the tea plant are as yet unknown. Here, two new PPO genes, CsPPO1 and CsPPO2, both of which had high identity with PPOs from other plants, were obtained from tea leaves. The fu… Show more

“…The expression vector carrying the jrPPO1-wt gene was transformed into E. coli BL21 (DE3) and was expressed similar to previous studies 21,39 , resulting in a final yield of 38 mg pure protein per liter of culture. However, a suitable protocol is required in order to avoid production of insoluble and/ or inactive PPO [40][41][42][43] . Different expression protocols have been reported, including the variation of affinity-tags, incubation temperatures and expression hosts (Table S2).…”

“…Polyphenol oxidase exhibits multiple important physiological effects such as resistance to stress, pests and diseases during growth and development ( Huang et al, 2018 ). During the manufacturing processes of black tea, the flavonoids present in the leaves are oxidized to o-quinones by PPO that are associated with the formation of pigments such as thearubigins, theaflavins, and theabrownin.…”

“… Zhang et al (2020) find that methyl jasmonate enhances PPO activity by activating the expression of CsPPO2 and CsPPO4 gene to improve defense against tea geometrid larvae ( Ectropis grisescens ) in tea plant. Additionally, mechanical damage or regurgitant of tea geometrid significantly upregulates the transcriptional expression of CsPPO1 and CsPPO2 ( Huang et al, 2018 ). These findings indicate that increasing the expression of PPO genes in tea plants is of interest; however, the molecular mechanism of the regulation of PPO activity in tea plants remains unclear.…”
