Two unrelated cell-derived sequences in the genome of avian leukemia and carcinoma inducing retrovirus MH2.

Abstract: Molecularly cloned proviral DNA of avian replication‐defective retrovirus Mill Hill No. 2 (MH2) was analyzed. The MH2 provirus measures 5.5 kb including two long terminal repeats (LTR), and contains a partial complement of the structural gene gag, 1.5 kb in size, near the 5′ terminus, and a 1.3‐kb segment of the v‐myc transforming gene near the 3′ terminus. These v‐myc sequences are closely related to the v‐myc transforming gene of avian acute leukemia virus MC29, and to the cellular chicken gene c‐myc. The ga… Show more

“…In contrast, transforming oncogenes induced anchorage-independent growth and abrogated contact inhibition. Cooperation of oncogenes was also observed in the study of viruses such as avian MH2 (17) or erythroblastosis (18) retroviruses. These experiments paved the way for the multistep theory of oncogenesis (10).…”

“…It was designated v-raf because of the ability of the virus to induce rapidly growing fibrosarcomas in mice (Rapp et al, 1983). Around the same time, chicken Raf was cloned from an avian virus (Mill-Hill No 2) and termed v-mil (Jansen et al, 1983(Jansen et al, , 1984. Cloning of human CRAF, ARAF and BRAF followed shortly afterwards (Bonner et al, 1985;Huebner et al, 1986;Beck et al, 1987;Ikawa et al, 1988;reviewed in Wellbrock et al, 2004).…”

“…2 MH2 was found to carry a second potential oncogene in addition to vmyc, which was termed v-mil, whereas its cellular counterpart was called c-mil. 3 Already a hybridization analysis and the gross comparison of restriction maps of v-raf and v-mil pointed at sequence homologies between these 2 genes. 4 By direct sequencing of both genes it was finally proven that 3611-MSV and MH2 have integrated orthologues of the same gene into their genomes.…”
