Ubp43 gene expression is required for normal Isg15 expression and fetal development

Abstract: Background: Isg15 covalently modifies murine endometrial proteins in response to early pregnancy. Isg15 can also be severed from targeted proteins by a specific protease called Ubp43 (Usp18). Mice lacking Ubp43 (null) form increased conjugated Isg15 in response to interferon. The Isg15 system has not been examined in chorioallantoic placenta (CP) or mesometrial (MM) components of implantation sites beyond 9.5 days post coitum (dpc). It was hypothesized that deletion of Ubp43 would cause disregulation of Isg15 … Show more

“…Usp18 (ubiquitin specific peptidase 18, also known as Ubp43), is an Isg15-specific protease that is up-regulated in response to interferon or lipopolysaccaride. Its gene expression is required for normal Isg15 expression (Rempel et al 2007). Oasl2 (2 0 -5 0 oligoadenylate synthetase-like 2), the third most predominantly up-regulated gene in the duodenum of Car6 -/-mice, is an allergy/inflammationrelated gene belonging to the 2 0 -5 0 oligoadenylate synthetase family, which was one of the first characterized IFN-induced antiviral proteins (Eskildsen et al 2003).…”

“…USP18 are believed to play essential roles in many biological functions, such as host innate immune response (Randall et al, 2006), brain cell injury , fetal development (Rempel et al, 2007), pancreatic beta cell inflammation and apoptosis (Santin et al, 2012) and so on. Recently the role of mammalian USP18 in viral infections was most comprehensively studied, indicating that this role is mediated both via its ISG15 protease action and in proteaseindependent fashions (Chen et al, 2011).…”

“…Furthermore, a recent study showed that Isg15 is expressed in both the antimesometrial plus mesometrial decidua and placenta on Days 12.5 and 17.5 of murine pregnancy, after decidualization is complete (Rempel and others, '07). Therefore, ISG15 likely also plays a role in the post-decidualization period.…”

“…Therefore, ISG15 likely also plays a role in the post-decidualization period. Although Isg15 knockout mice appear to have no problems during pregnancy (Osiak and others, '05), it has been suggested this observation may depend on their genetic background (Rempel and others, '07). Regardless, it seems that Isg15 is not absolutely required for successful pregnancy.…”
