Ultrahigh‐performance liquid chromatography coupled with triple quadrupole and time‐of‐flight mass spectrometry for the screening and identification of the main flavonoids and their metabolites in rats after oral administration of Cirsium japonicum DC. extract

Abstract: The metabolic pathways of pectolinarin, linarin and pectolinarigenin were summarized. This study not only proposed a practical strategy for rapidly screening and identifying metabolites but also provided useful information for further pharmacological studies and the design of new drugs based on Cirsium japonicum DC.

“…14 In recent years, high-performance liquid chromatography coupled with mass spectrometry (HPLC-MS/MS) has been routinely used to determine the contents of chemical compounds in traditional chemical materials and also applied in pharmacokinetic studies in rats and humans. 15,16 Moreover, it has been used to study drug metabolism, [17][18][19][20][21][22] toxicokinetics, 23,24 lipidomics, 25,26 proteomics 27,28 and metabolomics. 29,30 The main advantage of mass spectrometry (MS/MS) is the ability to detect a broad range of drugs with high sensitivity and specicity in a single analytical run.…”

“…The characteristic fragment ion at m / z 505.3441, m / z 481.3888, and m / z 463.3781 signified that phosphorylation occurred at the parent nucleus. Therefore, M9 was identified as the phosphorylation metabolite of RPDQ [ 28 ].…”

“…Further investigation suggests that pectolinarin, a flavonoid compound, may serve as the key active component in Cirsium japonicum responsible for promoting scarless regeneration. 8 Pectolinarin has garnered increasing attention due to its hemostatic, anti-inflammatory, antibacterial, antiviral and other pharmacological activities, and has been utilized in the treatment of various conditions such as diabetes and tumours. [9][10][11] Moreover, pectolinarin has shown promise in the field of tissue repair, where its antioxidant properties have been beneficial in treating liver damage.…”
