Unsuspected findings about phylogeny and ultrastructure of the enigmatic cyanobacterium Microcrocis geminata resulted in its epitypification and novel placement in Geminocystaceae

Abstract: The genus Microcrocis belongs to long ago established cyanobacterial taxa for which relevant data required for a modern taxonomic analysis remained unknown for a long time. In present study we proposed an epitype originating from a large natural population of the type species M. geminata. Through isolating clean individual colonies from the environmental sample, sufficient material for a single colony PCR and amplification of genomic DNA by MDA method was obtained. Molecular analyses of the 16S rRNA gene deter… Show more

“…Additional strains. CALU 1334, CALU 1794, CALU phenotypically identified as M. geminata demonstrated corresponding similarities of 16S rRNA gene and the ITS region to those in the family Geminocystaceae (Pokorný et al 2023, Strunecký et al 2023. Importantly, the morphology of M. geminata has been described using only environmental samples.…”

“…At present, the family Geminocysta ceae encompasses six genera characterized to varying degrees. For example, although the genus Microcrocis (see below) encompasses many phenotypically described representatives, only one species has been genotyped, and cultured strains have not been reported (Pokorný et al 2023).…”

“…The morphology of coccoid cyanobacteria is poorly characterized, compromising their taxonomical classification, which was until recently based mainly on morphological characteristics [11,12]. In recent years, a significant effort to improve the coccoid cyanobacteria taxonomy has been made using a polyphasic approach with molecular and morphological data for the description of new taxa or its reassessment [13,14], and more recently using genomic data [11,15].…”
