Dro1/Ccdc80 has been identified as a tumor suppressor gene in colorectal, thyroid, and ovarian cancer. Dro1/ Ccdc80 is ubiquitously expressed and has, next to its tumor suppressive role, been implicated in a wide range of biological processes, including adipogenesis, lens development, and skeletogenesis. DRO1/CCDC80 is a secreted protein that assembles with the extracellular matrix and promotes cell adhesion. The biological mechanisms by which DRO1/CCDC80 exerts its various functions are still widely unknown. DRO1/ CCDC80 inhibits malignant growth properties of cancer cells, mediates sensitization to apoptosis, regulates E-cadherin expression, and acts as a binding partner for Janus kinase 2 and cyclic GMP-dependent protein kinase 1. Moreover, DRO1 has been linked to MEK/ERK, fibroblast growth factor (FGF), Hedgehog, and Wnt/β-catenin signaling. In the following, DRO1/CCDC80 is presented as a novel tumor suppressor of colorectal carcinogenesis.

Androgen exposure stimulates the growth of beard hair follicles. The follicle dermal papilla appears to be the site of androgen action; however, the molecular mechanisms that regulate this process are not well understood. In an attempt to identify genes that contribute to the androgen-responsive phenotype, we compared gene expression patterns in unstimulated and androgen-treated cultured human dermal papilla cells isolated from beard (androgen-sensitive) and occipital scalp (androgen-insensitive) hair follicles. Through this analysis, we identified three genes that are expressed at significantly higher levels in beard dermal papilla cells. One of these genes, sfrp-2 has been identified as a dermal papilla signature gene in mouse pelage follicles. Two of these genes, mn1 and atp1beta1, have not been studied in the hair follicle. A fourth, fibulin-1d, was slightly upregulated in beard dermal papilla cells. The differences in the expression of these genes in cultured beard and scalp dermal papilla cells reflected similar differences in microdissected dermal papilla isolated from intact beard and scalp follicles. Our findings introduce potentially novel signaling pathways in dermal papilla cells. In addition, this study supports that cultured dermal papilla cells provide a cell-based model system that is reflective of the biology of in vivo hair follicle cells.

In agreement, AIB1 represses DRO1 promoter and its expression levels inversely correlate with DRO1 in several cancer cell lines and in ectopic and silencing assays. Estrogen modulators treatment showed a regulation in an estrogen receptor-dependent fashion. Importantly, DRO1 overexpression resulted in BCLAF1 upregulation, a compelling concept given that BCLAF1 is a death-promoting transcriptional repressor. Additionally, DRO1 shuttles from Golgi to the endoplasmic reticulum upon apoptotic stimuli, where it is predicted to facilitate the apoptosis cascade. Finally, DRO1 repression is an important factor for AIB1-mediated inhibition of apoptosis. Collectively, our results reveal DRO1 as an AIB1-targeted tumor suppressor, providing a novel mechanism for AIB1-dependent inhibition of apoptosis.
