Genetic relationships among 125 Spanish melon ( Cucumis melo L.) accessions from a Spanish germplasm collection were assessed using a standard molecular-marker array consisting of 34 random amplified polymorphic DNA (RAPD) markers bands (19 primers) and 72 reference accessions drawn from previous studies. The reference accession array consisted of a broad range [Japanese (19) Crete (17), African (15), and USA and Europe (US/EU, 21)] of horticultural groupings (Group Cantalupensis, Group Conomon, Group Inodorus, Group Flexuosus, and Group Chito), and of melon market classes (e.g., Charentais, U.S. Western and European Shipper types, Ogen, and Galia, Honeydew, and Casaba). Spanish melon accessions (largely Casaba, Group Inodorus) were genetically distinct from the reference accessions and other Group Inodorus melons of different origins. Most African accessions showed common genetic affinities, and grouped with the Group Chito and the Group Conomon accessions examined. Those accession groupings were distinct from all other accessions belonging to Group Cantalupensis, Flexuosus, and Inodorus accessions originating from Crete, Japan, Europe, and the U.S. Genetic diversity was highest in accessions of African origin and lowest in accessions of Spanish origin. Additional RAPD markers (49 primers, 141 bands) and 22 selected agronomic traits (quantitative and qualitative) were then used to assess the genetic diversity among Spanish accessions. While cluster analysis using fruit characteristics grouped accessions into cultivars, RAPD-based genetic-distance estimate did not provide consistent accession groupings either by cultivar or geographic origin. While the highest level of polymorphism was detected among melons originating from the central region of Spain, and in the Rochet cultivar, accessions from the Andalucía region and Green cultivars were comparatively less diverse. These results indicate that the Spanish melon accessions could be used to broaden the genetic base of local and foreign Casaba germplasm, to enhance the genetic diversity of U.S and European commercial melon germplasm, and to delineate collection strategies for acquisition of additional Spanish landraces.

The genetic diversity and relationship among South and East Asian melon Cucumis melo L. were studied by using RAPD analysis of 69 accessions of melon from India, Myanmar, China, Korea, and Japan. The genetic diversity was large in India, and quite small in Group Conomon var. makuwa and var. conomon from East Asia, clearly indicating a decrease in genetic variation from India toward the east. Cluster analysis based on genetic distance classified 17 groups of accessions into two major clusters: cluster I comprising 12 groups of accessions from India and Myanmar and cluster II that included five groups of accessions of Group Conomon var. makuwa and var. conomon from East Asia. Cluster I was further divided into three subclusters, of which subclusters Ib and Ic included small-and large-seed type populations, respectively. Therefore, this division was based on their seed size, not cultivation area. The large-seed type from east India was differently included in the subcluster of smallseed type (Ib). A total of 122 plants of 69 accessions were classified into three major clusters and subclusters: clusters I and II comprised melon accessions mostly from India and Myanmar, and cluster III comprised Group Conomon var. makuwa and var. conomon from East Asia. The frequency of large-and small-seed types was different between clusters I and II, also indicating genetic differentiation between large-and small-seed types. One plant of the small-seed type from east India was differently included in cluster III, and two plants from east India were classified into subcluster IV. These results clearly showed that South Asian melon is genetically differentiated by their seed size, and that small-seed type melon in east India is closely related to Group Conomon var. makuwa and var. conomon.

Genetic relatedness and phenotype are important factors that govern the expression of heterosis in hybrid progeny of many cross-pollinating plant species. Since this relationship is important but not well understood in melon (Cucumis melo L.), one monoecious and two andromonoecious melon lines of diverse Chinese [Peoples Republic of China (PRC)] origin were crossed to the andromonoecious U.S. Western Shipping market type 'Top Mark' (TM) and the andromonoecious, highly branched line H-16 to determine parental combining ability and heterosis for five yield component traits in three test environments [open-field (USA), and energy-saving (PRC) and plastic greenhouses (PRC)]. Random amplified polymorphic DNA (RAPD)-and simple sequence repeat (SSR)-based genetic distances (GD) among and between parents (5) and their hybrids (6) were calculated and compared to phenotypic trait values. These germplasms were evaluated for lateral branch number (LBN), days to 50% flower (DF), fruit number and weight per plant, and fruit length:diameter (L:D) ratio in each of three test environments. General combining ability was significant for all characters, except for L:D in all locations, and LBN and DF in the plastic greenhouse environment. Both descriptors of difference (genetic marker and phenotype) were discriminatory, and provided similar assessments of relationships among parents and hybrids. Although dramatic performance differences were detected between parents and among F 1 hybrid progeny, a strong relationship between GD and heterotic effects was not consistently detected.
