Variable lymphocyte receptors in hagfish

Abstract: A previously uncharacterized type of variable lymphocyte receptors (VLR) was identified recently in the Sea lamprey. This jawless vertebrate generates an extensive VLR repertoire through differential insertion of neighboring diverse leucine-rich repeat (LRR) cassettes into an incomplete germ-line VLR gene. We report here VLR homologs from two additional lamprey species and the presence of two types of VLR genes in hagfish, the only other order of contemporary jawless vertebrates. As in the Sea lamprey, the inc… Show more

“…The germline-diversified V domains in NITRs are an example of an adaptive immune feature in an innate immune receptor 73,74 , whereas germline joining of immunoglobulin-gene clusters in cartilaginous fish is an example of an innate immune feature in an adaptive immune receptor 18 . The Dscam variants in insects 10 , the FREPs in snails 8,126 , the VLRs in the jawless vertebrates lampreys 9 and hagfish 99 , and the VCBPs in the protochordate amphioxus 7,113 all share certain fundamental characteristics with the adaptive antigen-binding immune receptors of jawed vertebrates.…”

“…Single-cell PCR is consistent with haplotypic exclusion, a characteristic of lymphocyte-mediated immunity and a central feature of clonal selection in RAG-mediated adaptive immunity. VLRs that undergo similar genetic rearrangement have also been characterized in hagfish 99 .…”

“…is comprised of clonally diverse lymphocytes that express variable lymphocyte receptors (VLRs) created by the combinatorial assembly of leucine-rich repeat (LRR) gene segments rather than the Ig V, D, and J gene segments used by jawed vertebrates (gnathostomes) (1,2). Two VLR genes, VLR-A and VLR-B, have been identified in both lamprey and hagfish (1)(2)(3).…”

“…Two VLR genes, VLR-A and VLR-B, have been identified in both lamprey and hagfish (1)(2)(3). The germline VLR genes are incomplete in that they have coding regions only for the invariant N-terminal and C-terminal sequences, which are separated by intervening sequences, lacking canonical splice sites (1,2).…”
