Variants of Elisa Method for Detection of the Carnation Mottle Virus

Abstract

All possibilities for staЬle diagпostic of stroпg iпfectioп апd ecoпomicaly importaпt for Carпatioп mottle virus have Ьееп explored. Comparative tests оп the seпsitivity of some different variaпts of ELISA have Ьееп carried out -direct апd iпdirect Ьу Clark апd Adams (1), Lommel et al. (6) апd Edwards апd Cooper (!!). There are used alkalшe phosphatase апd peroxidase coпjugates. The coпditioпs for the differeпt tests to Ье carried out are optimized -coпceпtratioпs of immuпoglobuliпs (IgG) апd coпз"ugates, the duratioпs of iпcubatioп of aпtibody апd aпtigeпe. The comparative observatioп on ELISA variaпts shows that DAS ELISA is the most sensitive -1 ngjml Ьу usiпg the peroxidase coпjugate. Carпatioп mottle и·rus {CaMV) сап Ье ideпtified in maximum diluted sap -1:500 000, as Ьу alkaliпe phosphatase апd peroxidase coпjugate as well. The secoпd is preferaЬle because of its low cost. The indirect variants are with less seпsitivity. Ву the method of Lommel et al. {6) 10 пgjml сап Ье detected and that quaпtity of virus is ideпtified Ьу two coпjugates eпough all right. Ву the method of Elwards g.пd Cooper (2} 1 ngjml сап Ье detected Ьу usiпg aпtisera iпstead of IgG апd long time of substrate iпcubation -16 hours. On the base of experimental series carred out for practical applications iп the process of productioп of healthy carпation plants DAS ELISA with peroxidase coпjugate is recommeпdiЬle as the most seпsitive апd effective method.
